Glycogen synthase kinase 3 (GSK-3) is a serine/threonine protein kinase that mediates the addition of phosphate molecules onto serine and threonine amino acid residues. First discovered in 1980 as a regulatory kinase for its namesake, glycogen synthase (GS), GSK-3 has since been identified as a protein kinase for over 100 different proteins in a variety of different pathways.

Findings suggest that GYS2 serves as a prognostic factor and functions as a tumor suppressor in HCC. The newly identified HBx/GYS2/p53 axis is responsible for the deregulation of glycogen metabolism.
Glycogen synthase kinase-3 (GSK3) is a proline-directed serine-threonine kinase that was initially identified as a phosphorylating and inactivating glycogen synthase (see GYS1, 138570).Two isoforms, alpha (GSK3A; 606784) and beta, show a high degree of amino acid homology (Stambolic and Woodgett, 1994).GSK3B is involved in energy metabolism, neuronal cell development, and body pattern Glycogen synthase catalyzes the rate-limiting step of glycogen biosynthesis and has two major isoforms in mammals -- muscle isoform (GYS1) and liver isoform (GYS2) respectively (1). Glycogen synthase kinase-3α (GSK-3α) and glycogen synthase kinase-3β (GSK-3β) phosphorylate glycogen synthase at multiple sites in its C-terminus (Ser641, Ser645, Ser649 and Ser653) inhibiting its activity (2, 3). 2001-04-01 Bachrach BE, Weinstein DA, Orho-Melander M, et al. When in the non-phosphorylated state, glycogen synthase does not require glucose-6-phosphate as an allosteric activator; when phosphorylated it does (By similarity). By similarity 2020-07-06 Glycogen synthesis is primarily regulated by modulating the activity of glycogen synthase. This enzyme exists in two forms, dephosphorylated (active or a) and phosphorylated (inactive or b ). It is regulated by covalent modification, in an inverse direction to that of glycogen phosphorylase. Glycogen synthase is an enzyme that is responsible in glycogen synthesis. It is activated by glucose 6-phosphate (G6P), and inhibited by glycogen synthase kinases ( GSK3 ). Those two mechanisms play an important role in glycogen metabolism.

Glucose 6-phosphate activates glycogen synthase, resulting in an active form of glycogen synthase b. Glycogen synthase, as discussed earlier, catalyzes the rate-limiting step in glycogen synthesis in the liver and in skeletal muscle, namely, the transfer of glucose monomers from UDP-glucose to the terminal branch of the growing glycogen chain via the formation of α(1→4) glycosidic bonds. 79 Several glycogen synthase isoforms exist – one specific to skeletal muscle (encoded by GYS1), and 2020-07-06 glycogen synthase: , glycogen starch synthase a glucosyltransferase catalyzing the incorporation of d -glucose from UDP- d -glucose into 1,4-α- d -glucosyl chains. Die Glykogensynthase ist das Schlüsselenzym der Glykogensynthese und damit das Enzym, welches der Regulation dieses Stoffwechselweges dient. The stimulation of glycogen-targeted protein phosphatase 1 (PP1), glycogen synthase, and glycogen synthesis by insulin was examined during the differentiation of 3T3-L1 fibroblasts into adipocytes. Insulin treatment barely changed the low levels of glycogen synthesis measured in fibroblasts. Following differentiation into adipocytes, insulin increased glycogen synthesis up to 40-fold. After Glycogen synthase kinase-3 (GSK-3) is a protein-serine kinase implicated in the hormonal control of several regulatory proteins including glycogen synthase and the transcription factor c-jun.
